General description
Mucin-2 (UniProt: Q02817; also known as MUC-2, Intestinal Mucin-2) is encoded by the MUC2 (also known as SMUC) gene (Gene ID: 4583) in human. Mucin-2 (MUC2) is a secreted protein that coats the epithelia of the intestines, airways, and other mucus membrane-containing organs. It provides a protective, lubricating barrier against particles and infectious agents at mucosal surfaces. Major constituent of both the inner and outer mucus layers of the colon and may play a role in excluding bacteria from the inner mucus layer. Mucin-2 is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to produce the mature form. It exists as a disulfide-linked homotrimer that can be highly O-glycosylated. It contains four VWFD domains and two VWFC domains. Mucin-2 undergoes proteolytic cleavage in the outer mucus layer of the colon, contributing to the expanded volume and loose nature of this layer that allows for bacterial colonization in contrast to the inner mucus layer which is dense and devoid of bacteria. The numerous O-glycans on the Mucin-2 are reported to serve as nutrients for the bacteria, serve as attachment sites, and probably contribute to the selection of the species-specific colon flora. Reduced levels of Mucin-2 have been linked to the early stage of colorectal cancer development, which is caused by methylation of the MUC2 promoter.
