General description

母亲抗decapentaplegic同源物2（UniProt Q15796；也称为hMAD-2，hSMAD2，JV18-1，MAD同源物2，Mad相关蛋白2，母亲抗DPP同源物2，SMAD 2，SMAD家族成员2，Smad2）由人的SMAD2（也称为MADH2，MADR2）基因（基因ID 4087）编码。Smad2是通过转化生长因子TGF-β和激活素1型受体激酶激活的受体调节的细胞内信号转换器和转录调节剂。响应于TGF-β信号，smad2通过其与受体激活（SARA）蛋白的SMAD锚的相互作用而被募集到活化的TGF-β受体，其中它被TGF-β受体磷酸化。磷酸化导致其从SARA分离，并与另一个家族成员smad4结合。然后，smad2-smad4复合物易位到细胞核中，在细胞核中它与靶启动子结合并与其他辅因子形成转录阻遏物复合物。Smad2也可以被激活素1型受体激酶磷酸化，从而转导激活素依赖性信号传导事件。
