一般描述
BCA蛋白测定基于双缩脲作用，即在碱性溶液中蛋白质将Cu2+还原为Cu+，并具有浓度依赖性地检测单价铜离子。二辛可宁酸是一种螯合还原铜的显色剂，产生紫色络合物，在562nm处有很强的吸光度（Smith1985，Wiechelman 1988）。该方法可用于多种样品的蛋白质浓度定量，可在几分钟内进行。
Novagen® BCA蛋白质测定试剂盒可用于在标准测定或微量测定配置中确定20-2000 µg/mL范围内的蛋白质浓度。试剂盒组分足以完成500个标准大小的反应（50 µl蛋白质样品加1 ml试剂）或2500个微量反应（25 µl蛋白质样品加200 µl试剂）。为了方便可靠地制备标准曲线，提供了BSA标准品（2 mg/mL）。
该测定法具有耐用性，可以在许多化合物存在下进行。某些试剂，包括螯合剂、强酸或碱和还原剂，会干扰该测定所依赖的还原和螯合反应（Brown 1989）。BCA测定与以下Novagen蛋白质提取和裂解试剂相容：BugBuster蛋白质提取试剂、PopCulture试剂、CytoBuster蛋白质提取试剂、Reportasol提取缓冲液和Insect PopCulture试剂。试剂盒文献中描述了去除或稀释干扰物质的选项。
