His•Bind树脂是一种不带电荷的IDA琼脂糖树脂。结合力约为8 mg/ml床体积。His•Bind树脂非常适合使用重力流柱或分批方法进行的中小型规模的纯化。
应用
His•Bind®树脂已用于：通过金属螯合层析纯化His•Tag®融合蛋白；用于纯化细菌产生的OCRL1WT/VAR磷酸酶结构域；研究Lowe综合征的异质性；在蛋白纯化中研究粪便梭菌对厌氧间苯三酚的降解；分离His6标记蛋白；用于研究人抑制因子激活蛋白1（RAP1）和胶质纤维酸性蛋白（GFAPɛ）蛋白在酵母模型系统中增加分泌酶活性的作用。
生化/生理作用
His•Bind®树脂有助于通过金属螯合色谱法纯化含有his标记序列的蛋白质。它利用His•Bind®缓冲试剂盒中提供的电荷缓冲液，将His标记的序列与固定在His•Bind®树脂上的Ni2+或Co2+阳离子有效结合。目的蛋白可以通过咪唑洗脱很容易地回收。His•Bind®树脂被设计为可再生和重复使用多次，使其成为一种经济高效的解决方案。它的多功能性允许在温和，非变性条件下或在胍或尿素存在的情况下进行蛋白质纯化，这取决于纯化过程的具体要求。
