抗iNOS免疫细胞化学的推荐程序

:1.在适当的培养基中培养细胞。

2.通过用含有以下成分的新鲜培养基预处理细胞16小时来诱导NOS:不添加(对照),干扰素-γ(50 ng/ml),LPS(30 µg/ml)或干扰素加LPS。

3.用磷酸盐缓冲盐水(PBS)清洗细胞3-5分钟。

4.在室温下用2%皂苷渗透10分钟。

5.室温下在4%福尔马林中固定10分钟。

6.用PBS清洗3-5分钟。

7.在室温下用PBS中以1:500稀释的抗iNOS培养20分钟。

8.用PBS清洗3-5分钟。

9.用生物素化二抗(即生物素结合的山羊抗兔IgG,目录号401313)培养。清洗;与抗生物素蛋白结合的碱性磷酸酶(目录号189726或189732)一起培养。

10.用萘酚AS磷酸盐/固红紫色LB(目录号477758)或萘酚AS磷酸盐/新品红(目录号477759)展开。
