对每批、每次的全部ChIPAb+抗体的染色质沉淀进行单独验证。每个ChIPAb+抗体组均包含对照引物（通过qPCR对每个批次进行检测），以在基因座特异性环境下对IP结果进行生物学验证。提供了qPCR方案和引物序列，使研究人员能够在染色质环境中使用我们的抗体时验证ChIP方案。每组还包括阴性对照抗体，以确保ChIP反应的特异性。
ChIPAb+CREB，抗体/引物组包括CREB抗体，阴性对照抗体（纯化的兔IgG）和qPCR引物，可扩增人cFos基因CRE侧翼的64 bp区域。CREB和阴性对照培养上清液以可扩展的“每个ChIP”反应大小提供，可用于功能上验证与CREB相关染色质的沉淀。

使用10 µL阴性上清液或CREB抗体和Magna ChIP® A试剂盒（目录号17-610），对从293T细胞制备的超声染色质（每IP 5 X 106细胞当量）进行染色质免疫沉淀。
使用cFos下游4Kb（阴性）和cFos CRE（阳性）通过qPCR验证了CREB相关DNA片段的成功免疫沉淀（图2）。 数据表示为每个IP样品相对于每个扩增子和ChIP样品的输入染色质的输入百分比。

代表性批次数据。
