一般描述

EGFR（表皮生长因子受体，ErbB-1）是一种受体酪氨酸激酶（RTK），它是受体酪氨酸激酶的EGFR/ErbB家族的四种成员之一。 EGFR 在基本的正常细胞过程的调节以及增生性疾病病（如癌症）的病理生理学中起关键作用。EGFR信号传导通路的激活与细胞增殖的增加、血管生成、转移和减少的细胞凋亡有关。通过结合配体，EGFR由无活性的单体形式转化为活性的二聚体。除了形成同型二聚体外，EGFR还可以与ErbB受体家族的另一个成员（例如ErbB2/Her2/neu）配对，以产生活化的异二聚体。EGFR二聚体化刺激其固有的细胞内蛋白酪氨酸激酶活性，这种活性导致EGFR的C末端结构域中Y992、Y1045、Y1068、Y1148 和Y1173位点上五个酪氨酸（Y）残基的受体酪氨酸激酶的胞浆结构域内的酪氨酸残基的转磷酸化和自磷酸化。 这种自磷酸化导致多个下游信号转导级联的激活，特别是通过Ras、 PI3激酶的 MAPK、Akt 和JNK通路和PLC通路，这些通路最终导致各种下游通路的激活。在激活过程中Tyr1069磷酸化。
