Wnt家族由参与细胞发育，去向决定，器官发生和肿瘤发生的配体组成。Wnt信号的信息已从果蝇中无翅（wg）信号的遗传分析中推断出来。当Wnt与其受体，卷曲蛋白（Fz）跨膜受体家族的成员以及脂蛋白受体–相关蛋白5和6（LRP-5/6）相互作用时，Wnt信号级联反应在细胞膜上启动。这种相互作用将信号中继到细胞内部，在那里disheveled（dsh）被带到膜上并被磷酸化/激活。Dsh促进GSK3激酶活性的失活，从而防止β-连环蛋白的磷酸化并使其积累，因为它避免了泛素化和随后的蛋白体降解。稳定的β-连环蛋白在细胞质中积累，在那里它与淋巴增强因子/T细胞因子（LEF/TCF）相互作用并转移到细胞核中。这导致TCF反应性基因的激活，该基因被认为在发育和癌症中起关键作用。该途径的负调节剂可能包括axin和肿瘤抑制因子APC。
