General description
44 kDa
SIRT3 is a member of the class I sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. SIRT3 is located in the mitochondria and has been implicated in regulating metabolic processes. Endogenous SIRT3 is a soluble protein located in the mitochondrial matrix. Overexpression of Sirt3 in cultured cells increases respiration and decreases the production of reactive oxygen species. Fasting increases Sirt3 expression in white and brown adipose tissue. A longer form of SIRT3 is present in the nucleus where it exhibits histone deacetylase activity. SIRT3 protects cardiomyocytes from stress mediated cell death due to deacetylation of a nuclear factor, Ku-70.
Immunogen
Epitope: C-terminus
The immunogen was a peptide corresponding the C-terminal region of human SIRT3.
Application
Anti-SIRT3 (C-terminus) Antibody is a Rabbit Polyclonal Antibody for detection of SIRT3 (C-terminus) also known sirtuin 3 & has been validated in WB, ELISA & IHC.

This antibody recognizes SIRT3. Endogenous levels of this protein vary among cell lines; the protein was easily detected in RAW264.7 and C2C12 cells but only weakly detected in HeLa cells, and not detected in Jurkat, HEK293 or PC3 cells.

Preparation Note
Stable for 6 months at 2-8°C from date of receipt. For long term storage, store at -20ºC. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Moderate mitochondrial stress can lead to persistent activation of cytoprotective mechanisms - a phenomenon termed mitohormesis. Here, we show that mitohormesis primes a subpopulation of cancer cells to basally upregulate mitochondrial stress responses, such as the mitochondrial unfolded protein response (UPRmt)
