General description
140 kDa
TrkA，表观分子量140 kDa，是一种高亲和力神经生长因子（NGF）。Trk原癌基因家族包含四个成员TrkA，TrkB，TrkC和TrkE，它们在整个中枢神经系统和周围神经系统中可变表达。TrkA与神经生长因子（NGF）结合并在酪氨酸残基（Tyr490，Tyr674，Tyr675，Tyr751和Tyr785）上自磷酸化以激活多个下游效应蛋白。Shc缔合和随后激活Ras-MAP激酶信号级联反应需要Tyr490处的磷酸化，从而激活Elk-1依赖性基因转录和神经突生长。Tyr674和Tyr675处的磷酸化位于TrkA酪氨酸激酶的催化域内，反映了Trk激酶的活性。另外，PI3-激酶缔合和Akt信号级联反应的激活需要Tyr751的磷酸化。
