一般描述

CREB是一种β-ZIP转录因子，可通过cAMP反应元件激活靶基因。CREB能够介导来自多种生理刺激的信号，从而调节多种细胞反应。虽然CREB在许多组织中表达，但它在神经系统中起着重要的调节作用。CREB被认为在促进某些神经元群体的神经元存活，前体增殖，神经突向外生长和神经元分化中起关键作用。CREB作为神经营养蛋白途径的介体促进生长和分化。CREB能够通过与不同的二聚化伴侣的相互作用选择性激活许多下游基因。CREB通过各种信号传导途径（包括Erk，Ca2+和应激信号传导）在Ser133处磷酸化而被激活。
仅在Ser133（~43 kDa）磷酸化后检测到CREB。可能识别ATF-1（〜38 kDa）和CREM（〜30 kDa），因为这两种蛋白与磷酸化免疫肽具有显著同源性。

应用

免疫沉淀：
10 μL先前批次已对用50 μM福司可林处理的500 μg细胞中的磷酸化VASP进行了免疫沉淀。

免疫细胞化学：
先前批次的1:1000稀释液在福尔马林固定石蜡包埋的正常和缺血大鼠脑切片中检测到磷酸化CREB。

凝胶位移（EMSA）：
独立实验室已证明该抗体发生凝胶转移。
使用抗磷酸化CREB（Ser133）抗体（兔多克隆抗体）已在EMSA、IHC、IHC（P）、IP、WB中高度发布和验证，以检测磷酸化-CREB（Ser133），也称为活性转录因子CREB、cAMP反应元件结合蛋白1。

识别p43磷酸化CREB。也可识别p30和p38 kDa蛋白。P30 & p38蛋白可以分别是磷酸化的CREM和ATF-1，因为这两种蛋白与磷酸化的CREB免疫肽具有显著的同源性。

形式：纯化
纯化的兔IgG，溶于含有0.1 M Tris-甘氨酸（pH 7.4）、15 M NaCl、0.05％叠氮化钠和30%甘油的缓冲液中。
通过蛋白A-琼脂糖纯化，然后使用未磷酸化的CREB亲和凝胶柱进行免疫吸附层析。

制备说明

自收到之日起，在-20°C条件下可稳定保存1年。

处理建议：
收到后，在取下瓶盖之前，将小瓶离心并轻轻混合溶液。分装至微量离心管中，储存于-20°C
。避免反复冻融循环，以免损坏IgG和影响产品性能。

分析说明

对照
福司可林刺激的WI-38核提取物。
已通过蛋白质印迹对用50 mM福司可林处理的小鼠NIH/3T3细胞的裂解液进行了评估。

蛋白质印迹分析：当通过SDS-PAGE在福司可林处理的NIH/3T3裂解液中以1:1,000的稀释度在Ser133上磷酸化时，该抗体检测到CREB，并转移至PVDF（Immobilon-P）。
