General description

Polo样激酶1（PLK-1）是一种丝氨酸-苏氨酸蛋白激酶，在细胞周期的M期起着多种重要功能，包括调节中心体成熟和纺锤体组装、去除染色体臂（chromosome arm）的粘连蛋白、APC/C 抑制剂的失活、有丝分裂结束和胞质分裂的调节。 表达的PLK-1 蛋白在G2和M 期聚集至最高水平，在有丝分裂以后和整个G1期下降至几乎不可测的水平，然后在S期再次开始聚集。

Immunogen

对应于人Polo样激酶1（PLK1）的6His标记全长融合蛋白。也称丝氨酸-苏氨酸蛋白激酶PLK。

Application

免疫沉淀：一个独立实验室报道，一个先前批次的该抗体可用于PLK1的免疫沉淀。

免疫组织/免疫细胞化学： 一个独立实验室报道，1 μg/mL的先前批次的该抗体在多聚甲醛处理、Triton X-100 固定的细胞中显示出PLK1 的阳性免疫染色。

Analysis Note

对照
阳性抗原对照：货号12-305，3T3/A31裂解液。加入2.5 μL 2-巯基乙醇/100 μL裂解液，煮沸5分钟以还原样本。在微凝胶每个泳道上样20 μg经还原的裂解液。
已通过蛋白质印迹对3T3/A31细胞的RIPA裂解液进行了评估。

蛋白质印迹分析：0.5-2 μg/mL该抗体在3T3/A31细胞的RIPA裂解液中检测到PLK1。
