General description
51 kDa（α）或46 kDa（β）
糖原合成酶激酶3（GSK-3）是一种多功能丝氨酸/苏氨酸激酶，它是各种信号通路的关键调节因子。据报道在哺乳动物中有GSK-3 的两个亚型：51 kDa 的GSK-3a和47 kDa 的GSK-3b。这两种亚型的激酶结构域有98%左右的同源性，但是最后76个C末端氨基酸残基只有36%左右相同。GSK-3a 的N末端含有富含甘氨酸的延伸区。研究人员也鉴定出一个GSK-3b的次要（占总量的~15%）剪接变体GSK-3b2，它的激酶结构域内含有一个13个残基的插入片段（insert）。与 ‘未剪接’ 的GSK-3b相比，它对 tau蛋白的激酶活性下降。 与同样存在于神经元过程的未剪接GSK-3b不同，GSK-3b2 主要位于神经元胞体中。GSK-3b 在Wnt 信号通路中起关键的抑制作用。Wnt基因编码分泌性富含半胱氨酸的大蛋白家族，该蛋白对于成年组织的发育和维持很重要。据报道Wnt 信号通路异常促进人类退化性疾病和癌症。
