人真核翻译延长因子1α有两种同工型，α1和α2。α1同工型基因编码延伸因子1复合物的一个亚基，该复合物负责将氨酰tRNA酶促递送至核糖体。α1同工型在脑、胎盘、肺、肝、肾和胰腺中表达。另一种同工型（α2）在脑、心脏和骨骼肌中表达。
在66%的Felty综合征患者中，α1同工型被确定为自身抗原。研究发现，α1同工型的基因在许多染色体上有多个拷贝，其中一些（如果不是全部）代表不同的假基因。α2的基因可能在卵巢癌的进展中起关键作用。

免疫原

使用钙调蛋白亲和层析从布氏锥虫中分离出的天然钙调蛋白结合蛋白。克隆CBP-KK1。

应用

免疫沉淀：
4 µg先前批次可免疫沉淀A431 RIPA细胞裂解液中的EF1α。
可使用这种经验证可用于IP & WB的EF1α抗体（克隆CBP-KK1）检测EF1α。

该抗体可识别EF-1α（延伸因子1α），相对分子量为53 kDa。偶尔，在A431细胞裂解液中也会检测到38kDa的蛋白。

形式：纯化
纯化的小鼠单克隆IgG1κ，溶于含0.1 M Tris-甘氨酸（pH 7.4）、0.15 M NaCl和0.05％叠氮化钠的缓冲液中。

制备说明

自收到之日起，在 -20°C 条件下可稳定保存 1 年。

处理建议：
收到后，在取下瓶盖之前，将小瓶离心并轻轻混合溶液。分装到微量离心管中，并储存于-20ºC。避免反复冻融循环，以免损坏IgG和影响产品性能。

分析说明

对未刺激的A431细胞、人Jurkat或小鼠3T3/A31的RIPA裂解液进行了常规评估。

蛋白质印迹分析：
0.5-2 µg/mL的该批次在未刺激的A431细胞RIPA裂解液中检测到EF1α。先前批次在人Jurkat和小鼠3T3/A31 RIPA细胞裂解液中检测到EF1α。
对照
阳性抗原对照：货号12-301，未刺激的A431细胞裂解液。加入2.5µL 2-巯基乙醇/100µL裂解液，煮沸5分钟以减少制备量。在微凝胶每个泳道上样20µg经还原的裂解液。
