General description

CLEC9A, also known as Dendritic cell natural killer lectin group receptor 1 (DNGR-1), and encoded by the gene Clec9a/Dngr-1, functions as an endocytic receptor on a small subset of myeloid cells specialized for the uptake and processing of material from dead cells. CLEC9A functions as a SYK-coupled C-type lectin receptor to mediate sensing of necrosis by the principal dendritic-cell subset involved in regulating cross-priming to cell-associated antigens. CLEC9A recognizes filamentous form of actin in association with particular actin-binding domains of cytoskeletal proteins, including spectrin, exposed when cell membranes are damaged, and mediates the cross-presentation of dead-cell associated antigens in a Syk-dependent manner. CLEC9A is localized to the plasma membrane. CLEC9A is expressed in dendritic cells and has multiple isoforms; isoform 4 is expressed at high levels by CD8+ dendritic cells and at low levels by plasmacytoid DCs but not by other hematopoietic cells.

A novel dendritic cell (DC)-restricted molecule, Clec9A, was identified by gene expression profiling of mouse DC subtypes. Based on sequence similarity, a human ortholog was identified. Clec9A encodes a type II membrane protein with a single extracellular C-type lectin domain.
