General description

蛋白质和脂质磷酸酶PTEN（10号染色体上缺失的磷酸酶和紧张素同源物）在肿瘤发生中起作用。PTEN是一种肿瘤抑制因子，其缺失可通过PI3K途径发出组成信号，并足以导致癌症。在正常细胞中，与FOXO1A和PTEN共同作用抑制细胞生长。受体激活后，PTEN代谢PI3K磷酸化PI(4,5)P2的产物，产生PI(3,4,5)P3。在缺乏PTEN的细胞中，PI(3,4,5)P3水平升高，它是促进肿瘤发生的有效第二信使。PTEN功能的恢复被认为是一种新的癌症治疗方法。过表达PTEN抑制细胞迁移，表达反义PTEN mRNA促进细胞迁移。
