General description

Imprint™ Methylated DNA Quantification technology provides a rapid and reliable method to measure global DNA methylation shifts. The Imprint Methylated DNA Quantification Kit contains all reagents required to detect relative levels of methylated DNA. Up to 200 ng of purified DNA is bound to the wells of the assay strip. The methylated DNA is detected using the Capture and Detection antibodies, then quantified colorimetrically. The amount of methylated DNA present in the sample is proportional to the absorbance measured.

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   6. Can I use chloroform to extract my DNA before using Product MDQ1, Imprint® Methylated DNA Quantification Kit?

    Choroform extraction should not interfere with the protocol as long as the DNA was not eluted with the chloroform.

  7. When using Product MDQ1, Imprint® Methylated DNA Quantification Kit, do I need to use all the wells in the plate at once?

    No, you can use as many wells as needed.  You will want to ensure that all the wells that are not being used are covered well with Parafilm, Saran Wrap or some other plate sealer.

  8. What is the detection limit of Product MDQ1, Imprint® Methylated DNA Quantification Kit?

    The detection limit of this kit is 5 nanograms of fully methylated DNA.

  9. When using Product MDQ1, Imprint® Methylated DNA Quantification Kit, can I dilute the entire antibody at once and save it for later?

    No. A fresh dilution of the antibody should be prepared each time the kit is used. Diluting the antibody all at once will decrease its stability and it's effectiveness in the kit.

  10. Are there any species restrictions for Product MDQ1, Imprint® Methylated DNA Quantification Kit?  

    The antibody works on 5-me-C, so any species with DNA that is methylated at the 5 position of cytidine residues will work. 

  11. Is Product MDQ1, Imprint® Methylated DNA Quantification Kit, shipped in two parts?

    Yes, Product MDQ1, Imprint® Methylated DNA Quantification Kit, is shipped in 2 parts. Part 1 contains, 2-8 °C components (W4267, D8568, C3493, D8818, D8693, S3447, and A4231) and Part 2 contains -20 °C components (M8570 and B6561).
