肿瘤坏死因子-α也称为恶病质素,表达为 26 kDa 的膜结合蛋白,然后被 TNF-α 裂解转化酶 (TACE) 释放可溶性 17 kDa 单体,在循环中形成同源三聚体。TNF-α在抗肿瘤活性、免疫调节、炎症、厌食、恶病质、感染性休克、病毒复制和造血中发挥作用。TNF-α对许多转化细胞具有细胞毒性,但在正常二倍体细胞中,可刺激增殖(成纤维细胞)、分化(骨髓细胞)或活化(中性粒细胞)。TNF-α 还显示出对 DNA 和 RNA 病毒的抗病毒作用,并诱导其他几种细胞因子的产生。
