一般描述

PCR DIG探针合成试剂盒包含碱不稳定性洋地黄毒苷 (DIG)-11脱氧尿苷三磷酸(dUTP)制剂。该试剂盒可方便有效地制备聚合酶链式反应(PCR)所需的高灵敏度DIG标记DNA探针。这一探针可通过PCR方法利用DIG-11dUTP（碱不稳定性）标记。DIG-dUTP:dTTP的比例为1:2。
PCR DIG探针合成试剂盒包含增强高保真DNA聚合酶混合物。这种强大的酶混合物具有校对活性，以10 pg质粒DNA和10 ng基因组DNA为模板，可聚合40 bp至5 kb长的探针。DIG标记探针可稳定储存一年以上。

应用

利用PCR DIG探针合成试剂盒制备的洋地黄毒苷(DIG)标记DNA探针适于检测Southern和Northern杂交中的低（单）拷贝基因mRNA。[1][2]DIG标记DNA探针也已用于原位杂交过程中的DNA标记。[3]
可根据探针长度调节提供的dUTP-核苷酸混合物浓度。可在琼脂糖凝胶上快速确定标记效果。
可按照包装说明书中的方案，多次抗体剥离和重新检测。
一次PCR标记反应 (50 μl)制备的探针，通常足够用于20 ml杂交溶液。本试剂盒可进行约25次反应(50 μl)。

质量

每个批次的PCR DIG探针合成试剂盒都在PCR中经过功能呢检测。扩增产品在基因组Southern blot中进行了检测。
在如本包装内插页所描述的PCR条件下，对照反应可生成442bp的扩增产物。由于在PCR过程中DIG-dUTP的多次插入，PCR产物的分子量将会相对于未标记PCR产物略有提高。在将PCR产物杂交至10μg人类基因组DNA后可通过后续的化学发光检测检测到特异性的片段模式。
