General description

2′-Deoxycytidine 5′-triphosphate (dCTP) is synthesized either by de novo pathway or from the multiple phosphorylation steps of cytidine.[1] It is deaminated to deoxyuridine monophosphate (dUMP) which is convertible to deoxythymidine triphosphate (dTTP).[1]

Application

2′-Deoxycytidine 5′-triphosphate disodium salt has been used:
  • in polymerase chain reaction mix for the amplification of Alternaria (AM) toxin DNA[2]
  • to spike oligonucleotide sample for capillary electrophoresis (CE) analysis[3]
  • as a component in adenosine 5′-monophosphate (AMP) luciferase assay[4]

2′-Deoxycytidine 5′-triphosphate disodium salt is one of the nucleoside triphosphates used for DNA polymerase driven reactions such as polymerase chain reaction, DNA sequencing and other molecular biology methods.

Biochem/physiol Actions

2′-Deoxycytidine 5′-triphosphate (dCTP) is used to synthesize DNA by DNA polymerases[5] or reverse transcriptases. dCTP is an allosteric regulator of deoxycytidylate (dCMP) deaminase.[6] It is used in studies of nucleotide pools, nucleotide selectivity and enzyme specificity studies, such as the preferred incorporation of dCTP by Rev1, a γ-family DNA polymerase.[7]
