General description

Liposomes contain a self-quenching fluorescent dye called calcein. It is a fluorescent dye with excitation and emission wavelengths of 495/515 nm, respectively. It self-quenches at concentrations above 70mM and is commonly used as an indicator of lipid vesicle leakage. The leakage rate is calculated on the basis of the fluorescence increment as the entrapped calcein is leaked out of the liposomal compartment. It is used as a complexometric indicator for titration of calcium ions with EDTA, and for fluorometric determination of calcium.[1][2]

Application

Calcein-AM has been used in flow cytometry.[3]
Non-fluorescent cell permeable derivative of calcein, becomes fluorescent on hydrolysis. Used as a neutral substrate for multidrug resistance protein, MRP; employed in tumor research.

      Calcein-AM is a cell-permeant and non-fluorescent compound that is widely used for determining cell viability. In live cells the non-fluorescent calcein-AM is hydrolyzed by intracellular esterases into the strongly green fluorescent anion calcein. The fluorescent calcein is well-retained in the cytoplasm in live cells.
