General description

Propidium iodide solution is used to assess neuronal death.[1]

Application

Membrane impermeant, phenanthridinium intercalator dye excited by mercury or xenon-arc lamps or argon-ion laser. Suitable for use in fluorescence microscopy, confocal laser scanning microscopy, flow cytometry, and fluorometry. Used as a nuclear counterstain.
Propidium iodide solution has been used:
  • in staining DNA[2]
  • in the viability staining method to study islet viability[3]
  • to visualize F-actins and nuclei in mesenchymal stem cell sheets[4]

      Propidium Iodide does not generally enter living cells,  However it may be taken up by axonal terminals and retrogradely transported to neuronal cell bodies. Reference: Aschoff, A. and H. Hollander, Fluorescent compounds as retrograde tracers compared with horseradish peroxidase (HRP). I. A parametric study in the central visual system of the albino rat. J. Neurosci. Methods, 6(3), 179-197 (1982).

    4. What are the excitation and emission maxima for Propidium Iodide, Product P4864?

      Excitation maximum is at 493 nm in water.  Emission maximum is at 636 nm in water

    5. How is Propidium Iodide, Product P4864, used for Flow Cytometry?

      Immediately before flow cytometric analysis, prepare a 0.5 mg/mL solution of propidium iodide by combining 50μL of 2X PBS with 50 μL of P4864. Add this solution to 1 mL of a cell suspension containing 10,000,000 cells. (Note: Cells exhibiting fluorescence above 630 nm should be excluded from further analysis.)
