一般描述

将 C18 柱方法转换至 Discovery® HS F5 柱的指导原则:
通常,碱性化合物在 HS F5 色谱柱上的保留时间比在 C18 柱上更长。在 C18 柱上流动相中的有机相含量提高 5% 至 10% 通常可获得与 HS F5 柱类似的保留值。使用其他化合物所得的结果差异则很大。然而 log Po/w 值小于 2.5 的化合物在 HS F5 柱上的保留时间通常要长于 C18 柱。保留时间的差异程度与化合物的 logP o/w 值密切相关。
Discovery® HS F5 键合相可提供与 C18 色谱柱截然不同的反相分离。由于 F5 色谱柱上各种化合物通常会在与 C18 相同的时间范围内洗脱出来,使得大部分的 C18 方法可轻松转换到 F5 上。
