General description

Histopaque-1077 is a sterile, endotoxin tested, ready-to-use medium. The Histopaque composition includes polysucrose and sodium diatrizoate, adjusted to a density of 1.077g/mL. This ready-to-use medium facilitates rapid recovery of viable lymphocytes and other mononuclear cells from small volumes of whole blood or bone marrow.

Application

Histopaque-1077 has been used for:
  • human lymphocyte antigen (HLA) typing
  • initial isolation step prior to the enumeration of T, B, and ‘null’ lymphocytes.
  • preparation of pure lymphocyte suspensions for cell culture and cytotoxicity assays.
  • isolation and purification of peripheral blood mononuclear cells (PBMCs) and bone marrow mononuclear cells (BMMCs)
  • isolation of human lymphocytes
  • isolation of breast cancer circulating tumor cells (CTCs) from the whole blood
  • Histopaque-1077 is used in conjunction with Histopaque-1119 as it facilitates the rapid recovery of viable mononuclear cells and granulocytes from small volumes of whole blood

Principle

Anticoagulated blood is layered onto Histopaque-1077. During centrifugation, erythrocytes are aggregated by polysucrose and rapidly sediment. Granulocytes become slightly hypertonic, which increases their sedimentation rate, resulting in pelleting at the bottom of the centrifuge tube. Lymphocytes and other mononuclear cells remain at the plasma/ Histopaque-1077 interface.

  1. Is Histopaque®, Product 10771, the same as Ficoll® solution, Product F5415?

    Ficoll-Hypaque is a mixture of Ficoll (polysucrose) and Hypaque (sodium diatrizoate).  It does not specify a density.  Product No. 10771 is a mixture of polysucrose and sodium diatrizoate at a density of 1.077 g/ml.

  2. Should I use Histopaque® 10771 or 10831 for isolation of lymphocytes?

    The different product numbers reflect the density of the solution.  Product No. 10771 is to be used to isolate human mononuclear cells.  Product No. 10831 is to be used for the isolation of mononuclear cells from mice, rats and other mammals.

  3. What is the difference between Histopaque® Product No. 10771 and H8889?

    Product 10771, Histopaque®-1077, is not cell culture tested and Product H8889, Histopaque®-1077 Hybri-Max, is cell culture tested.Both Products are sterile, endotoxin tested and intended for Research Use Only. 

  4. What is the osmolarity of the Product 10771, Histopaque®-1077 solution? 

    The osmolarity is a property that is tested for each lot of material; the range of acceptable values is 280.0 - 300.0 mOs/kg.  The lot specific value is given on the Certificate of Analysis.  Certificates of Analysis can be viewed on our website by entering the lot number on our Advanced Search Page.
