General description

SIGMAFAST Protease Inhibitor Cocktail Tablets, EDTA-Free is a water-soluble tablet formulation designed to inhibit serine, cysteine, aspartic and metalloproteases. The tablets contain no strong metal chelators, such as EDTA, and are compatible with sample processing by immobilized metal affinity chromatography (IMAC). The tablets also contain a unique, proprietary formulation that includes a pepstatin A salt for additional protection.

Application

The broad specificity of SIGMAFAST Protease Inhibitor Cocktail Tablets makes them suitable for use in the purification of histidine-tagged proteins and as a general-purpose protease inhibitor cocktail. The tablets can also be used as components of buffers for cell lysis, and have been optimized and tested for use with HIS-Select® media.

Features and Benefits

EDTA-Free: Contains no strong metal chelators, such as EDTA, which might interfere with IMAC processing.

Pepstatin A: Contains a unique, proprietary formulation that includes a pepstatin A salt for additional protection not normally obtained in a tablet format.

Broad Specificity: Inhibits serine, cysteine, aspartic, and metalloproteases, providing broad protection for protein samples.

High Yield: One tablet makes 100 mL of cocktail, and is recommended for the inhibition of the proteases present in a maximum of 20 g of cell mass in 100 mL of extract.

Long Stability: The tablets are stable as supplied for at least 2 years at 2–8 °C, and the reconstituted protease inhibitor solution (1´ or 10´) is stable for at least 2 weeks at 2–8 °C.

Quantity

Storage and Stability

  5. What is the difference between Protease Inhibitors P8849 and S8830?

    P8849 is a DMSO solution and S8830 is a water soluble tablet. P8849 formulation is considered proprietary but does contain AEBSF, Pepstatin A, Bestatin, E-64, and Phosphoramidon. The formulation of S8830 also contains Aprotinin and Pepstatin A and the exact formulation is given on the product information sheet (under Documents, above).

  6. Can Product No. P8849, Protease Inhibitor Cocktail, be used with with HIS-Select products or any other immobilized metal affinity chromatography (IMAC) products?

    This product does not contain EDTA and can be used with sample processing by immobilized metal affinity chromatography (IMAC) suchas used for the purification of histidine-tagged proteins..

  7. How should I dissolve Product No. S8830, SIGMAFAST Protease Inhibitor?

    You can dissolve the product in 100 ml of water to make a 1X solution. You can make a 10X stock solution using 10 ml of water and then dilute 1:10 for use.  Concentrations greater than 1X may appear slightly hazy. This will not affect the performance of the protease inhibitors.

  8. Are there publications that cite use of Product S8830, SIGMAFAST Protease Inhibitor Cocktail Tablets?

    Yes. Several examples of published papers that cite the use of Product S8830 include these:1.  Smith, E.R., et al., Instability of fibroblast growth factor-23 (FGF-23): Implications for clinical studies.  Clinica Chimica Acta, 412(11-12), 1008-1011 (2011).  Use was with blood at 20 μL cocktail per 1 mL of blood.2.  Reber, S., "Isolation of Centrosomes from Cultured Cells" (Chapter 8), from Microtubule Dynamics: Methods in Molecular Biology (Springer Protocols), vol. 777, pp. 107-116 (2011).
