General description

SIGMAFAST Protease Inhibitor Tablets are a water-soluble tablet formulation containing a broad-spectrum cocktail of protease inhibitors, including AEBSF, E-64, bestatin, leupeptin, aprotinin, and EDTA. One tablet can make 100 mL of cocktail and can inhibit proteases present in up to 20 g of cell extract.
The use of SIGMAFAST Protease Inhibitor Tablets is not advised with HIS-Select® products or other immobilized metal affinity chromatography (IMAC) products. These tablets contain EDTA, which is a metal chelator and can potentially remove metal ions from these affinity products.

Specificity

Inhibits serine, cysteine, aspartic, and metalloproteases.

Application

SIGMAFAST Protease Inhibitor Tablets has been used:
  • to wash raw semen for membrane protein analysis
  • in a solution to incubate CD4+ T cells to obtain total cell lysates for western blot analyses
  • as a component of NP-40 buffer to homogenize the spinal cord tissue centered at the injury epicenter to conduct western blot analysis
  • as a component in radio-immunoprecipitation assay (RIPA) buffer to homogenize the spinal cord tissue centered at the injury center to detect neuregulin-1 beta 1 (Nrg-1ß1) in spinal cord tissue samples with enzyme-linked immunosorbent assay (ELISA)

These tablets have been optimized and tested for general use. They have been used in various applications, including collecting tracheae in combination with ice-cold radioimmunoprecipitation assay buffer to determine the tracheal basal cell fate, and homogenizing rat lung tissues for western blot analysis.

Quantity

One tablet makes 100 mL of cocktail. One mL is recommended for the inhibition of proteases equivalent to 1 mg of USP pancreatin. One tablet is recommended for the inhibition of proteases present in a maximum of 20 g of cell extract.

    S8820 is the tablet version of P2714.  

  6. Can Product S8820, SIGMAFAST Protease Inhibitor Tablets, be used with with HIS-Select products or any other immobilized metal affinity chromatography (IMAC) products?

    The tablets contain EDTA, a metal chelator, which may remove metal ions from these affinity products. The suggested protease inhibitor for the HIS-Select products is either Product No. S8830 (water soluble tablet) or P8849 (DMSO solution).

  7. Can Product S8820, SIGMAFAST Protease Inhibitor Tablets, be used with ELISA?

    Yes, because it is water based.

  8. How should I dissolve Product S8820, SIGMAFAST Protease Inhibitor Tablets?

    You can dissolve the product in 10 ml of water to make a 10X stock solution and then dilute 1:10 for use.

  9. What is the concentration of the inhibitors in Product S8820, SIGMAFAST Protease Inhibitor Tablets?

    When reconstituted (one tablet in 100 ml water), concentrations of the component inhibitors are as follows:AEBSF      2 mME-64          14 μMBestatin     130 μMLeupeptin   0.9 μMAprotinin    0.3 μMEDTA        1 mM
