General description

Adenosine triphosphate (ATP) is a nucleoside triphosphate formed exclusively in the mitochondria. It is a high-energy molecule described as the energy currency in all living systems. The chemical energy contained in the phosphate bond of ATP drives most cellular processes.[1][2] Genetic disorders such as Leber hereditary optic neuropathy,[2] Leigh syndrome, neuropathy, ataxia, and retinitis pigmentosa [3] affect the generation of ATP in the mitochondria.

Application

ATP Assay Kit has been used to quantify the ATP concentration by fluorometric detection of ATP in samples.[4]

Suitability

Suitable for the determination of ATP in samples such as animal tissue and cell culture samples.

Principle

The ATP Colorimetric/Fluorometric Assay Kit is a sensitive assay for determining ATP in various samples (ranging from 2–10 nmole/well for the colorimetric assay and from 20–1,000 pmole/well for the fluorometric assay). ATP concentration is determined by phosphorylating glycerol, resulting in a colorimetric (570 nm) or fluorometric (lex = 535/lem = 587 nm) product proportional to the amount of ATP present.

Other Notes

The fluorometric assay is 10-100 times more sensitive than the colorimetric assay.
