Specificity

Clone 2N12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects DNA (cytosine-5)-methyltransferase 3B (DNMT3B). It targets an epitope within 22 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal region of human DNA (cytosine-5)-methyltransferase 3B (DNMT3B).

Application

Quality Control Testing

Evaluated by Western Blotting in Caco-2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected DNMT3B in Caco-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected DNMT3B in COS-7 and NTERA-2 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DNMT3B in Human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound DNMT3B peptide with a KD of 3.2 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A representative lot of this antibody bound DNMT3B in K562 cells.

Target description

DNA (cytosine-5)-methyltransferase 3B (UniProt: Q9UBC3; also known as EC:2.1.1.37, Dnmt3b, DNA methyltransferase HsaIIIB, DNA MTase HsaIIIB, M.HsaIIIB) is encoded by the DNMT3B gene (Gene ID: 1789) in human. DNMT3B is a ubiquitous protein that is highly expressed in fetal liver, heart, kidney, and placenta. In several other tissues it is expressed at lower levels. It is the major de novo DNA methyltransferase that is expressed and is active during the early stage of embryonic development, including implantation. It plays a key role in the control of sense-antisense gene expression and the splicing of alternative exons. In addition to its role to methylate centromeric, pericentromeric, and subtelomeric repeats, it also acts as the main enzyme methylating intragenic regions of active genes. DNMT3B shares a high degree of homology with DNMTA3 in its catalytic domain but displays distinct and overlapping functions with DNMT3A. DNMT3B is recruited to target sites in a tightly regulated manner that involves the crosstalk between the de novo methyltransferase, chromatin remodeling complexes, histone modifications, and transcription factors. DNMT3B contains a PWWP domain (aa 225-283) that is essential for targeting to pericentric heterochromatin. Mutations in DNMT3B gene are known to cause immunodeficiency-centromeric instability-facial anomalies syndrome 1 (ICF1), a rare disorder characterized by recurrent infections, facial anomalies, and branching of chromosomes. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
