Specificity

Clone 1C19 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Apoptosis-associated speck-like protein containing a CARD (ASC/PYCARD). It targets an epitope within 22 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human Apoptosis-associated speck-like protein containing a CARD (ASC/PYCARD).

Application

Quality Control Testing

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected ASC/PYCARD in THP-1 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected ASC/PYCARD in HL-60 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ASC/PYCARD in THP-1 cells.

Flow Cytometry Analysis: 1 μg from a representative lot detected ASC/PYCARD in one million THP-1 cells.

Affinity Binding Assay: A representative lot of this antibody bound ASC with a KD of 1.1 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ASC/PYCARD in human tonsil tissue sections.

Target description

Apoptosis-associated speck-like protein containing a CARD (UniProt: Q9ULZ3; also known as hASC, Caspase recruitment domain-containing protein 5, PYD and CARD domain-containing protein, Target of methylation-induced silencing 1) is encoded by the PYCARD (also known as ASC, CARD5, TMS1) gene (Gene ID: 29108) in human. PYCARD is widely expressed at low levels and is detected in peripheral blood leukocytes, lung, small intestine, spleen, thymus, and colon. At significantly reduced levels it is also found in placenta and liver, and kidney. It is detected in the nucleus, mitochondria, endoplasmic reticulum, cytoplasm, and Golgi apparatus. It is localized primarily to the nucleus in resting monocytes/macrophages and rapidly redistributed to the cytoplasm upon pathogen infection. PYCARD functions as key mediator in apoptosis and inflammation and promotes caspase-mediated apoptosis. It is involved in activation of the mitochondrial apoptotic pathway where it promotes caspase-8-dependent proteolytic maturation of BID independently of FADD in certain cell types and also mediates mitochondrial translocation of Bax and activates Bax-dependent apoptosis coupled to activation of caspase-9, -2 and -3. In innate immune response, it acts as an integral adapter in the assembly of the inflammasome that activates caspase-1 leading to processing and secretion of proinflammatory cytokines. Its function as an activating adapter in different types of inflammasomes is mediated by the pyrin (aa 1-91) and CARD (aa 107-195) domains and their homotypic interactions. It is shown to be essential for recruitment of caspase-1 to inflammasomes containing certain pattern recognition receptors, such as NLRP2, NLRP3, AIM2 and probably IFI16. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
