General description

Adenomatous polyposis coli protein (APC) is encoded by the gene mapped to human chromosome 5q. The encoded protein is composed of 2843 amino acids. APC exists as many transcripts and they are expressed in tissue specific manner.
This gene encodes a tumor suppressor protein that acts as an antagonist of the Wnt signaling pathway. It is also involved in other processes including cell migration and adhesion, transcriptional activation, and apoptosis. Defects in this gene cause familial adenomatous polyposis (FAP), an autosomal dominant pre-malignant disease that usually progresses to malignancy. Disease-associated mutations tend to be clustered in a small region designated the mutation cluster region (MCR) and result in a truncated protein product. (provided by RefSeq)

Biochem/physiol Actions

Adenomatous polyposis coli protein (APC) is a tumor suppressor gene. It plays an important role in cell division, adhesion and migration. Mutations in this gene are associated with familial adenomatous polyposis (FAP). APC is involved with Wnt pathway and inhibits the molecules which induce β-catenin expression, an agent participating in cellular transformation. It also regulates metaphase-anaphase transition, mitotic exit and G1 phase, thereby maintaining genomic stability and chromosome segregation.
