General description

CD54 (cluster of differentiation 54), also known as ICAM1 (intercellular adhesion molecule 1) was initially identified as a ligand for the β2 integrin lymphocyte function-associated antigen (LFA)-1. It is a glycoprotein with a 90,000-110,000 molecular weight and shows wide range of tissue expression, including tissue macrophages and dendritic cells. It also shows expression on non-hematopoietic cells such as vascular endothelial cells, epithelial cells of thymus and mucosa and dermal fibroblasts. It is a member of the ICAM family, which also includes ICAM-2, -3, -4 and -5.
The antibody 1H4 reacts with CD54 (ICAM-1), a 85-110 kDa type I transmembrane glycoprotein (receptor for rhinovirus) expressed on activated endothelial cells, T lymphocytes, B lymphocytes, monocytes, macrophages, granulocytes and dendritic cells; the expression of CD54 is upregulated by activation.

Application

The reagent is designed for Flow Cytometry analysis of human blood cells using 20 μL reagent / 100 μL of whole blood or 1e6 cells in a suspension. The content of a vial (2 mL) is sufficient for 100 tests.

Biochem/physiol Actions

The expression of CD54 (cluster of differentiation 54)/ICAM1 (intercellular adhesion molecule 1) is strongly increased by interleukin-1 (IL-1) and interferon (IFN)-γ. It is expressed on lymphoid cells where it is required for phorbol ester-induced aggregation. It participates in lymphocyte function-associated antigen 1 (LFA-1)-dependent NK cell stimulation through its attachment to actin cytoskeleton through α-actinin or ezrin. It also acts as the receptor for rhinovirus, a common cold virus and Plasmodium falciparum. Soluble ICAM-1 levels are elevated in pregnant women with gestational diabetes mellitus (GDM), which is an outcome of endothelial cell activation and dysfunction. Meta-analysis of case-control studies show that rs1799969 polymorphism in ICAM-1 gene is linked with increased susceptibility to cancer.
