General description

Tumor necrosis factor receptor 2 (TNFR2) gene with ten exons and nine introns, spanning approximately 43kbp is mapped to human chromosome 1p36.3-p36.2. TNFR2 is a member of TNF receptor superfamily, and it is expressed highly on cells of hematopoietic lineage. TNFR2, also known as CD120b or p75, TNF receptor is a 75-kDa type I transmembrane protein, characterized with cysteine-rich extracellular regions with three to six disulfide-linked domains required for ligand binding.

The antiserum was produced against synthesized peptide derived from human TNF Receptor II.

Application

Anti-TNF Receptor 2 antibody has been used to study the effect of TNF signalling on CPF- and CPF+ CM (pesticides)-induced toxicity or apoptosis in SH-SY5Y cells.

Biochem/physiol Actions

Tumor necrosis factor receptor 2 (TNFR2) plays a vital role in inducing signals for various cellular process, including proliferation of thymocytes and peripheral T cells, T cell secretion of granulocyte-macrophage colony-stimulating factor, inhibition of early hematopoiesis and the inactivation and clearance of TNF by the kidney. Mutation in the gene leads to hematopoietic cell malignancies, neuroblastoma, glioma and cervical and ovarian carcinoma. TRAF2 interacts with TNFR2 and activates NF-κB (NF-κB) pathway. TNFR2, which lack death domains modulates the effect of other death receptor TNFR1, which induces T cell death. TNFR2 plays an essential role in TNFα–induced leukocyte-endothelial interaction. TNFR2 is considered as a potential therapeutic alternative, as it has capacity to nullify the common side effects associated with drugs targeting TNF-α or p55.
