General description

MART-1 (melanoma antigen recognized by T cells) is also termed as Melan-A. It is a type III membrane protein with a molecular weight of 22-24 kDa. It is present in the endoplasmic reticulum (ER), trans-Golgi network (TGN) and in melanosomes. MART-1 is a melanocyte lineage protein. This gene is mapped to human chromosome 9p24.1.
Melanoma-associated antigen recognized by T cells-1 (MART-1) (also known as Melan-A) is a melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. It is a transmembrane protein which is hydrophobic in nature. Six other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, Tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. The protein is also expressed in melanoma tumors. Anti-MART-1 Antibody detects endogenous levels of total MART-1 protein.

Biochem/physiol Actions

MART-1 (melanoma antigen recognized by T cells) is essential for the activities of the melanosomal matrix protein PMEL17 (premelanosome protein)/GP100 and the maturation of melanosomes. It plays a key role in pigmentation. MART-1 plays an important role in the expression, stability, trafficking and processing of Pmel17.
Melanoma-associated antigen recognized by T cells-1 (MART-1) has been shown to be essential for the functioning of glycoprotein-100 (gp100).
