General description

Glutamate decarboxylase 2 (GAD2) encodes GAD65, an isoform of glutamate decarboxylase. GAD65 is majorly expressed in axon terminal regions in neurons. GAD65 is present in the synaptic vesicles. GAD2 is mapped to human chromosome 10p12.1. It has an N-terminal pyridoxal 5′-phosphate (PLP) binding −domain.

Application

Monoclonal Anti-GAD65 antibody produced in mouse has been used in the detection of recombinant GAD65 using ELISA.

Biochem/physiol Actions

Glutamate decarboxylase 2 (GAD2) protein GAD65 catalyzes the formation of γ-aminobutyric acid (GABA) from glutamic acid. Palmitoylation of GAD65 is crucial for protein sorting in synaptic clusters. GAD65 autoantigen is prevalent in type 1 diabetes. Its expression increases on allergen exposure. GAD65, along with γ-aminobutyric acid (GABA), may play a key role in the pathogenesis of asthma.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide
