Gene Information

zebrafish ... pcna(30678)

General description

Proliferating cell nuclear antigen (PCNA) is a ring-shaped homotrimer protein, that is located at the center of the faithful duplication of eukaryotic genomes. Since it encircles the DNA, PCNA is also known as a sliding clamp. This gene is mapped to human chromosome 20p12.3.

Immunogen

Recombinant protein containing a sequence corresponding to a region within amino acids 31 and 222 of pcna according to NP_571479

Application

Suggested starting dilutions are as follows: IHC-P: 1:100-1:1000, WB: 1:500-1:3000, IHC-Wm: 1:50-1:500, Immunostaining: Assay-dependent dilution. Not yet tested in other applications. Optimal working dilutions should be determined experimentally by the end user.
Anti-PCNA antibody has been used in the detection of proliferating cells by immunofluorescence.

Biochem/physiol Actions

This protein is an auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase′s processibility during elongation of the leading strand.
Proliferating cell nuclear antigen (PCNA) controls the production of the leading and lagging strands, which are required for the duplication of DNA. It acts as a cell cycle regulatory protein. This protein regulates apoptosis. PCNA is also involved in non-replicative DNA synthesis events.
