General description
The gene encoding arachidonate 5-lipoxygenase (ALOX5) is localized on human chromosome 10q11.21 and has 13 introns. It spans a region of 71.9 kbp. The ALOX5 protein consists of 673 amino acids and has a molecular weight of 77.9 kDa. It possesses two domains, a regulatory C2-like domain that binds to calcium and a carboxy-terminal catalytic domain. ALOX5 is expressed in B cells, dendritic cells, mast cells, macrophages and polymorphonuclear leukocytes.
This gene encodes a member of the lipoxygenase gene family and plays a dual role in the synthesis of leukotrienes from arachidonic acid. The encoded protein, which is expressed specifically in bone marrow-derived cells, catalyzes the conversion of arachidonic acid to 5(S)-hydroperoxy-6-trans-8,11,14-cis-eicosatetraenoic acid, and further to the allylic epoxide 5(S)-trans-7,9-trans-11,14-cis-eicosatetrenoic acid (leukotriene A4). Leukotrienes are important mediators of a number of inflammatory and allergic conditions. Mutations in the promoter region of this gene lead to a diminished response to antileukotriene drugs used in the treatment of asthma and may also be associated with atherosclerosis and several cancers. Alternatively spliced transcript variants have been observed, but their full-length nature has not been determined. (provided by RefSeq)

Biochem/physiol Actions
The gene ALOX5 (arachidonate 5-lipoxygenase) encodes a lipoxygenase that catalyzes the conversion of arachidonic acid (AA) and other fatty acids to bioactive compounds such as 5(S)-hydroxyeicosatetraenoic acid (5-HETE), leukotriene B4 (LTB4), and other leukotrienes. These leukotrienes and lipoxins function as mediators of inflammatory response. ALOX5 has been implicated in tumorigenesis and may serve as a potential therapeutic target and biomarker.
