General description

Pepsinogen II is an aspartic protease produced majorly in the gastric chief cells. It is broadly dispersed in the gastrointestinal tract of various species. Pepsinogen II is a seminal plasma protein, which also exists as a zymogen in gastric mucosa. It comprises chiefly of β-sheets with a substrate-binding cleft. The cleft is situated between the two lobes consisting of two catalytic-site aspartates (Asp32 and Asp215). Pepsinogen II gene is mapped to the human chromosome 6p21.1.

Biochem/physiol Actions

Pepsinogen II functions as a proteolytic enzyme of gastric secretion. It also exhibits aspartyl proteinase-like features. Pepsinogen II degrades dietary proteins in the stomach, whereas it stimulates defensins for local antimicrobial defense in seminal fluids. Pepsinogen II is critical in processing the secretory pathway of alveolar type-2 cells. It is also used as a marker in several neoplasms and cancers.
The ratio between serum levels of pepsinogens I and II can be determined to identify patients with high risk of gastric cancer.
Used as a marker of gastric mucosa status.
