Application

For assay of 3′,5′-cyclic nucleotides.

Biochem/physiol Actions

Hydrolyzes the 3′,5′-phosphodiester in cyclic nucleotide monophosphates, such as cAMP and cGMP, to the corresponding nucleotide 5′-monophosphate.

Quality

Contains inorganic pyrophosphatase and 5′-ATPase.
Substantially free of 5′-nucleotidase and 2′,3′-cyclic nucleotide phosphodiesterase.

Unit Definition

One unit will hydrolyze 1.0μmole of 3′,5′-cyclic-AMP to 5′-AMP per min at pH7.5 at 30°C.

Lyophilized preparation containing near saturation levels of both protein activator and Ca++, and buffer salts as imidazole and magnesium sulfate

Prepared as described by Butcher, R.W. and Sutherland, E.W., J. Biol. Chem., 237, 1244 (1962).

Reconstitution

Reconstitute with 50% glycerol. The total activated units of enzyme will remain constant for at least 5 days when stored at −0 °C. However, the activator deficient activity may increase up to 200%. Both the activated and activator deficient activity may decrease approx. 30% in 24 hrs. if stored at 4 °C.
