General description

Dulbecco′s Modified Eagle′s medium (DME) is a modification of Basal Medium Eagle (BME) that comprises a four-fold higher concentration of amino acids and vitamins, as well as additional supplementary components. The original DME formula, first reported for culturing embryonic mouse cells, contained 1,000 mg/L of glucose. An alteration with 4,500 mg/L glucose is optimal in cultivating certain cell types.

Application

Dulbecco′s Modified Eagle′s medium-low glucose has been used in the differentiation and culture of human and murine mesenchymal stem cells (MSCs).

Quantity

Formulated to contain 10.0 grams of powder per liter of medium.

Reconstitution

Supplement with 3.7 g/L sodium bicarbonate.

  4. How do I prepare Product D5523, Dulbecco’s Modified Eagle’s Medium - low glucose?

    Measure out 90% of final required volume of water. Water temperature should be 15-20 °C.While gently stirring the water, add the powdered medium. Stir until dissolved. Do NOT heat.Rinse original package with a small amount of water to remove all traces of powder. Add to solution in step 2.To the solution in step 3, add 3.7 g sodium bicarbonate or 49.3 ml of sodium bicarbonate solution [7.5%w/v] for each liter of final volume of medium being prepared. Stir until dissolved.While stirring, adjust the pH of the medium to 0.1-0.3 pH units below the desired pH since it may rise during filtration. The use of 1N HCl or 1N NaOH is recommended.Add additional water to bring the solution to final volume.Sterilize immediately by filtration using a membrane with a porosity of 0.22 microns.Aseptically dispense medium into sterile container.

  5. How do I store Product D5523, Dulbecco’s Modified Eagle’s Medium - low glucose, dry powder?  How do I store the reconstituted medium?

    Store the dry powdered medium at 2-8 °C under dry conditions and liquid medium at 2-8 °C in the dark.  The nature of supplements added may affect storage conditions and shelf life of the medium. The product label bears the lot-specific expiration date.

  6. What sodium bicarbonate should I use with Product D5523, Dulbecco’s Modified Eagle’s Medium - low glucose?

    Sodium Bicarbonate powder (Product No. S5761) or Sodium Bicarbonate Solution, 7.5% (Product No. S8761) is recommended.  For more information, consult the product datasheet.

  7. Do you offer a liquid version of Dulbecco’s Modified Eagle’s Medium - low glucose?

    Product No. D6046 is the fully supplemented liquid form of the original Dulbecco's formulation, offered as Product No. D5523, with one key exception.  Pyridoxal, which is an aldehyde, has been replaced with pyridoxine.  This modification has been shown to improve the stability of the medium.  D5546 is the same but does not contain L-glutamine.
