General description

DAPI is an indolenine dye. It carries two amidine components, one on a pendant phenyl ring and another directly on the indole moiety.

Application

DAPI, dilactate has been used for the nuclear staining in cells.[1][2] DAPI is a common flow cytometry stain that is widely used as a DNA and nuclear stain. Additionally, it is used as a counterstain following in situ hybridization and other immunofluorescence procedures.

Biochem/physiol Actions

A high sensitivity dye used to detect single nucleic acid molecules. DAPI has been shown to detect a single DNA molecule in solution, and to detect femtograms of DNA in single cells and chloroplasts.

  4. Which filter sets can be used to visualize the fluorophore produced by Product D9564, DAPI, dilactate (DAPI-DNA)?

One such filter set is the Zeiss Filter Set 01 (ex=359 nm, em=461 nm), information about which is posted here.

  5. Isn't the hydrochloride salt form of DAPI a more sensitive detector of DNA than Product D9564, DAPI, dilactate (DAPI-DNA)?

The dilactate salt has as much sensitivity in detection as the dihydrochloride salt, which we also offer as Product No. D9542.

  6. Can Product D9564, DAPI dilactate (DAPI-DNA), be used as a vital stain?

As we note in the Product Information Sheet for the Dihydrochloride salt of DAPI, Product No. D9542, DAPI can be used as a vital dye to stain mature pollen grains, but we have tested it as a vital stain in other cell types.
