RNA免疫沉淀(RIP)是研究核糖核蛋白(RNP)复合物中RNA结合蛋白(RBP)及其RNA靶点的强大方法。抗特定RBP的抗体被用于共沉淀RNP,即RBP及其RNA伴侣。然后,可以通过下一代测序鉴定RNA或通过RT-PCR检测特定RNA。
当与合适的抗体联用时,Imprint RNA免疫沉淀试剂盒可提供RIP实验成功所需的所有试剂。该试剂盒包括两种裂解缓冲液:一种较为温和,与RNP结合不紧密;另一种则很苛刻,可与RNP密切结合。其中随附蛋白A磁珠,用于收集免疫沉淀物,方便洗涤。该试剂盒还包括用于阴性对照RIP的兔和小鼠IgG,以及一种兔抗鼠的桥接抗体,用于将小鼠单克隆一抗有效结合至磁珠上的蛋白A;其中还含有蛋白酶和核糖核酸酶抑制剂,可保护RIP实验过程中RNP不被降解。
