该系统可简单特异地检测生理条件下活细胞内游离一氧化氮(NO)和一氧化氮合成酶(NOS)的活性。一氧化氮由L-精氨酸经NOS作用而形成,是一种独特的细胞信号分子,可同时作为细胞内和细胞外的信使分子。NO与血管舒张、神经传递、细胞毒性和炎症都有一定关联。
一氧化氮合酶活性的标准检测方法针对的是不含细胞的可溶性酶制品,如细胞裂解物、组织粗提物或纯化酶。该方法可检测由放射性标记的精氨酸底物转换而成的放射性瓜氨酸的产量(Sigma货号NOS-1)。与之不同的是,本品是基于荧光的细胞相关NOS检测系统,通过非放射性方法检测细胞内NO产量。 采用可穿过细胞膜的二乙酸盐形式的二氨基荧光素(DAF-2 DA)替代放射性同位素。DAF-2 DA可迅速穿入细胞,进入细胞后被细胞内酯酶水解成为DAF-2,DAF-2与经NOS作用产生的NO反应,生成带有荧光的三唑荧光素。随后,可在激发波长492 nm 和发射波长515 nm 下,对该荧光性产物进行定量检测。
