General description
Hemoglobin subunit alpha (UniProt: P69905; also known as Alpha-globin, Hemoglobin alpha chain) is encoded by the HBA1 (also known as HBA2) gene (Gene ID: 3039, 3040) in human. Hemoglobin subunit alpha is a critical component of the hemoglobin molecule, responsible for oxygen transport in red blood cells. It is a globular protein with eight alpha-helical segments that form a compact, stable structure. Each hemoglobin molecule is a tetramer consisting of two alpha (HBA1) and two beta subunits, coordinating iron-containing heme groups that reversibly bind oxygen. Hemoglobin subunit alpha is involved in both fetal hemoglobin (HbF, alpha-gamma composition) and adult hemoglobin (HbA, alpha-beta composition), ensuring oxygen delivery throughout different developmental stages. HBA1 is localized in erythrocytes and functions in essential biochemical pathways such as oxygen transport, carbon dioxide release, and buffering pH in blood. Mutations or deletions in the HBA1 gene are associated with disorders like alpha-thalassemia, where reduced or absent alpha-globin chain production leads to imbalanced globin chain synthesis. This condition manifests in varying severity, from asymptomatic carriers to life-threatening conditions like Hb Bart syndrome, characterized by severe anemia and hypoxia. Other pathologies include methemoglobinemia (excessive oxidation of heme iron) and erythrocytosis, which alters oxygen affinity. The interaction of HBA1 with beta-globin (HBB) subunits forms the basis of hemoglobin′s cooperative oxygen-binding mechanism, mediated by allosteric transitions between relaxed (R) and tense (T) states. Dysregulation of these mechanisms, as seen in hemoglobinopathies, contributes to tissue hypoxia, oxidative stress, and systemic complications. Recent studies also implicate alpha-globin in non-erythroid tissues, suggesting roles in oxidative metabolism and cellular protection. Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HBA1/HBA2 in human placenta tissue lysate.
Affinity Binding Assay: A representative lot of this antibody bound with HBA1/HBA2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected HBA1/HBA2 in human placenta tissue sections.
