Specificity

Clone 1O10 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects IL-10.

Immunogen

Full-length mature (without the signal peptide) recombinant human IL-10.

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant Human IL-10.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected full-length, mature, recombinant Human IL-10 (without the signal peptide).

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound recombinant Human IL-10 with a KD of 1.0 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:100 dilution from a representative lot detected IL-10 in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution o from a representative lot detected IL-10 in Jurkat cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected IL-10 in one million Human peripheral blood mononuclear cells (PBMC) treated with phorbol 12-myristate 13-acetate( PMA; 500 ng/mL), Ionomycin (500 ng/mL) and Brefeldin A (100 ng/mL) for 4- 6 hours.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-10 (UniProt: P22301; also known as IL-10, Cytokine synthesis inhibitory factor, CSIF) is encoded by the IL10 gene (Gene ID: 3586) in human. IL-10 is a homodimeric secreted anti-inflammatory cytokine produced by a variety of cells, including T-cells, macrophages, and mast cells.  It is a major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions.  It is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature form. IL-10 acts on many cells of the immune system to limit excessive tissue disruption caused by inflammation. During infection, it inhibits the activity of Th1 cells, NK cells, and macrophages, all of which are required for optimal pathogen clearance. It inhibits the release of pro-inflammatory cytokines.  It ameliorates the excessive Th1 and CD8+ T cell responses that are responsible for much of the immunopathology associated with infections. It is shown to control the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling. IL-10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB, which leads to JAK1 and STAT2-mediated phosphorylation of STAT3 that in turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
