Specificity

Clone 2B3 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects MMP-7. It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of Human MMP-7.

Application

Quality Control Testing

Evaluated by Western Blotting in HT-29 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected MMP-7 in HT-29 cell lysate.

Tested Applications

Enzyme Immunoassay (ELISA) Analysis: A serial of dilutions from a representative lot detected recombinant Human MMP-7.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MMP-7 in HT-29 cells.

Target description

Matrilysin (UniProt: P09237; also known as EC:3.4.24.23, Matrin, Matrix metalloproteinase-7, MMP-7, Pump-1 protease, Uterine metalloproteinase) is encoded by the MMP7 (also known as MPSL1, PUMP1) gene (Gene ID:  4316) in human. MMPs are a family zinc-dependent endoproteinases that degrade collagen and other structural proteins and are responsible for much of the turnover of matrix components. Each MMP contains a protease domain with a conserved HExxGHxxGxxHS/T sequence, where three histidine residues form a complex with a catalytic Zn atom.  They contain a generally conserved sequence PRCGVP around the chelating cysteine that is known as the “cysteine switch.” They are produced as zymogens, with a signal sequence and propeptide segment that must be removed during activation. The propeptide domain contains a conserved cysteine, which chelates the active zinc site. MMP-7 is a calcium and zinc-dependent endopeptidase that is synthesized with a signal peptide (aa 1-17) and a propeptide (aa 18-94) that are subsequently cleaved off to produce the active enzyme. It number of substrates including collagen types IV and X, elastin, fibronectin, gelatin, laminin, and proteoglycans. It is the smallest of all the MMPs and lacks the hemopexin-like domain that is common to other members of the family.  MMP-7 is widely expressed, and its elevated levels are observed in cycling endometrium, colorectal cancers and adenomas, hepatocellular carcinomas, pancreatic carcer, and in about 50% of gliomas. MMP-7 play a vital role in the maintenance of innate immunity in organs such as the lungs and intestines where it proteolytically activates anti-bacterial peptides such as pro-defensins. It is also required for mediating proteolytic release of TNF from macrophages. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
