Specificity
Clone 4M20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Ras GTPase-activating-like protein IQGAP1. It targets an epitope within 18 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal region of human Ras GTPase-activating-like protein IQGAP1.
Application
Quality Control Testing
Evaluated by Western Blotting in HEK293 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IQGAP1 in HEK293 cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IQGAP1 in CHO K1, COS-7, and A549 cell lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IQGAP1 in human colon tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound IQGAP1 with a KD of 5.6 x 10-7 in an affinity binding assay.

Target description
Ras GTPase-activating-like protein IQGAP1 (UniProt: P46940; also known as p195) is encoded by the IQGAP1 (also known as KIAA0051) gene (Gene ID: 8826) in human. IQGAP1 is a cell-cell contact regulator that also plays a crucial role in regulating the dynamics and assembly of the actin cytoskeleton. Its expression is reported in the placenta, lung, and kidney and low level of expression is observed in the heart, liver, skeletal muscle, and pancreas. In eukaryotic cells IQGAP1 binds to and alters the function of several proteins, including actin, E-cadherin, beta-catenin, Cdc42, and Rac1. It is localized in the nucleus, cytoplasm, and membrane and its subcellular distribution is regulated by the cell cycle. Its nuclear levels are reported to increase at the G1/S phase. It may also play a role in cell cycle regulation by contributing to cell cycle progression after DNA replication arrest. IQGAP1 contains a calponin homology domain (aa 44-159 that is responsible for actin binding, a WW motif (aa 679-712) that is essential for the association of ERK2, four calmodulin-binding IQ domains (aa 745-864), and a RasGAP-related domain (aa 1004-1237) that binds the small GTPases Cdc42 and Rac1. IQGAP1 is reported to inhibit GTPase activity of Cdc42 and overexpression of IQGAP1 in cells is shown to increase Cdc42-GTP levels. Two short C-terminal fragments of IQGAP1 (C1: aa 956-1274; and C2: 1276-1657) are shown to bind exclusively to nucleotide-depleted Cdc42 (Cdc42-ND), whereas the entire C-terminus (aa 956-1657) binds predominantly to the Cdc42-GTP S. Phosphorylation of IQGAP1 at Ser 1443 by protein kinase Cϵ is reported to inhibit the intramolecular interaction of the C-terminal fragments. Ser 1443 phosphorylation causes an opening of the IQGAP1 structure and exposes C1 and C2 regions. Ser 1443 phosphorylation also enhances the ability to promote neurite outgrowth. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
