Specificity

Clone 1J2-H1 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Galectin-3. It targets an epitope within 17 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal region of human Galectin-3.

Application

Quality Control Testing

Evaluated by Western Blotting in RAW264.7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Galectin-3 in RAW264.7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Galectin-3 in NIH3T3 and MDA-MB-231 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Galectin-3 in NIH3T3 and RAW264.7 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Galectin-3 in human small intestine tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Galectin-3 (UniProt: P17931; also known as Gal-3, 35 kDa lectin, CBP 35, Galactose-specific lectin 3, Galactoside-binding protein, GALBP, IgE-binding protein, L-31, Laminin-binding protein, Lectin L-29, Mac-2 antigen) is encoded by the LGALS3 (also known as MAC2) gene (Gene ID: 3958) in human. Galectins are a family of β-galactoside-binding lectins with an evolutionary conserved carbohydrate-recognition domain. Galectin-3, a galactose-specific lectin, is widely expressed and is found in immune cells, epithelial cells, endothelial cells, and sensory neurons. Its expression is developmentally regulated and is more abundant during embryogenesis and development.  It is predominantly located in the cytoplasm and shuttles into the nucleus. It is also secreted to the cell surface and into biological fluids. In the cytoplasm, galectin-3 plays a role in cell survival by interacting with certain survival-associated proteins, including Bcl-2 and GTP-bound K-Ras. In the nucleus, it promotes pre-mRNA splicing and regulates gene transcription. Extracellular galectin-3 is reported to modulate cell-cell interactions. Its secretion is dependent on protein unfolding and is facilitated by the cargo receptor TMED10. Galactin-3 binds proteins in a carbohydrate dependent and  independent manner. It is reported to bind with IgE and with α3/β1 integrin mediates endothelial cell migration. In association with DMBT1, it plays a role in the terminal differentiation of columnar epithelial cells during early embryogenesis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.03 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
