Specificity
Clone 1D23-1 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects P2X purinoceptor 7 (P2X7). It targets an epitope within 23 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal region of human P2X purinoceptor 7 (P2X7).
Application
Quality Control Testing
Evaluated by Western Blotting in Human brain tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected P2X7 in Human brain tissue lysate.
Tested Applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected P2X7 in SK-MEL-28 cells.
Enzyme Immunoassay (ELISA) Analysis: A serial of dilution from a representative lot detected GST-tagged full-length Human P2X7.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected P2X7 in Human cerebral cortex tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound P2X7 peptide with a KD of 2.0 x 10-9 in an affinity binding assay.

Target description
P2X purinoceptor 7 (UniProt: Q99572; also known as P2RX7, ATP receptor, P2Z receptor, Purinergic receptor) is encoded by the P2RX7 gene (Gene ID: 5027) in human. P2RX7 is a multi-pass membrane protein of the P2X receptor family that is widely expressed with highest levels reported in brain and immune tissues. It contains two cytoplasmic domains (aa 1-25; 356-595); two transmembrane domains (aa 26-46; 335-355); and an extracellular domain (aa 47-334). Eight different isoforms of P2RX7 have been described that are generated via alternative splicing. Functional P2RX7 are shown to be organized as homomeric and as heteromeric trimers. They ACT® as receptor for ATP and serve as a ligand-gated iOn™ channel. They are also responsible for ATP-dependent lysis of macrophages through the formation of membrane pores that are permeable to large molecules. They can function in both FAST synaptic transmission and in ATP-mediated lysis of antigen-presenting cells. P2RX7 can undergo phosphorylation that results in its inactivation. ADP-ribosylation at arginine 125 is shown to be essential for its channel gating activity. Also, palmitoylation of several cysteines in the C-terminal cytoplasmic tail is required for efficient localization to cell surface. In the absence of ATP, it can function as a scavenger receptor in the recognition and engulfment of apoptotic cells. P2X7 receptor is dephosphorylated at tyrosine 343 upon activation, which strongly inhibits the membrane current and retards membrane blebbing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
