Specificity

Clone 1E14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Tumor protein 63 (TP63). It targets an epitope within 13 amino acids from the N-terminal region. It is predicted to react with isoforms 2, 4, 8, and 12.

Immunogen

KLH-conjugated linear peptide corresponding to 13 amino acids from the N-terminal region of isoform 2 of human Tumor protein 63 (TP63).

Application

Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TP63 in A431 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TP63 in Mouse E18 skin tissue lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TP63 in A431 cells.

Affinity Binding Assay: A representative lot of this antibody bound TP63 peptide with a KD of 6.9 x 10-6 in an affinity binding assay.

Target description

Tumor protein 63 (UniProt: Q9H3D4; also known as p63, Chronic ulcerative stomatitis protein, CUSP, Keratinocyte transcription factor KET, Transformation-related protein 63, TP63, Tumor protein p73-like, p73L, P40, P51) is encoded by the TP63 (also known as KET, P63, P73H, P73L, TP73L) gene (Gene ID: 8626) in human. TP63 is a member of the p53 protein family that is widely expressed. It is structurally related to the p53, however its C-terminal region differs from p53′s in that it contains a sterile alpha motif (SAM) domain (aa 541-607) and is subject to multiple alternative splicings. It acts as a sequence specific DNA binding transcriptional activator or repressor. It binds to DNA as a homotetramer. Its DNA binding region is localized to amino acids 170-362. It has a transcription activation (TA) region that is localized in amino acids 1-107 and a transactivation inhibition region that resides in amino acids 610-680, which can interact with and inhibit the activity of the N-terminal TA domain of TA-type isoforms. Twelve different isoforms of TP63 have been described that are produced by alternative promoter usage and alternative splicing. These isoforms contain a varying set of transactivation and auto-regulating transactivation inhibiting domains thus showing an isoform specific activity. The ratio of DeltaN-type (those lacking the TA domain) and TA-type isoforms governs the maintenance of epithelial stem cell compartments and regulates the initiation of epithelial stratification from the undifferentiated embryonal ectoderm. Isoform 2 is reported to activate RIPK4 transcription and may be required in conjunction with TP73/p73 for initiation of p53/TP53 dependent apoptosis in response to genotoxic insults and the presence of activated oncogenes. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
