Specificity

Clone 1G21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects E. coli β-Galactosidase. It targets an epitope within 24 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal region of E.coli β-galactosidase.

Application

Quality Control Testing

Evaluated by Western Blotting with E. coli β-Galactosidase.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected E.coli β-Galactosidase.

Tested applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected β-Galactosidase in HeLa cells infected with Adenovirus expressing β-Galactosidase.

Enzyme Immunoassay Analysis (ELISA): A serial of dilutions from a representative lot detected E. coli β-Galactosidase.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Beta-galactosidase (UniProt: P00722; also known as EC:3.2.1.23; Beta-gal) is encoded by the LacZ gene (Gene ID: 945006) in E. coli. The β-galactosidase is a tetrameric enzyme composed of four polypeptide chains (A-D), each with 1023 amino acids. Each monomer contains five domains (1-5). Domain 3 has an /β or TIM barrel structure with the active site located at the C-terminal end of the barrel. The residues that form the active site are contributed by different segments of the polypeptide chain. β-galactosidase hydrolyzes the disaccharide lactose to generate galactose and glucose and can also convert lactose to allolactose, which is the natural inducer for the lac operon. It displays strict specificity at the galactosyl position but can hydrolyze β-D-galactopyranosides with a wide variety of aglycons. Its substrate binding site is localized to amino acids 538-571. The hydrolysis reaction is a double displacement reaction where the product retains the same stereochemistry as the starting state. In the initial step, the substrate forms a covalent -D-galactosyl enzyme intermediate with the nucleophile Glu537. In the second step the intermediate is released, a step that is facilitated by Glu461, which abstracts a proton from the acceptor molecule, R OH. The activity of β-galactosidase is dependent upon the presence of divalent (Mg2+) and monovalent (Na+) cations. Mg2+ causes a 5-to-100-fold activation, depending on the substrate. However, Mg2+ can be replaced by Mn2+ to obtain full activity. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
